Antimicrobial activity of lactoferrin
The antimicrobial effect of lactoferrin on microorganisms was evaluated according to the method described by Gao *et al*[[@B46]]. Briefly, 50 μl of *E. coli*and *S. aureus*suspension (1 × 10^6^CFU/ml) was added to 25 ml TSB in 125 ml serum bottles. Lactoferrin was added at various concentrations in triplicate, and cultures were incubated at 37°C for 24 h. Afterwards, samples were plated on blood agar plates, incubated for 24 h at 37°C and the total CFU was counted. The effect of lactoferrin on *L. monocytogenes*was tested by incubating the bacteria with different concentrations of lactoferrin. Cultures were incubated at 37°C in anaerobic jars for 48 h and then inoculated on BHI agar and the plates were incubated at 37°C for 24 h. The effect of lactoferrin on *L. monocytogenes*was also tested on blood agar plates under aerobic conditions.
Extraction of genomic DNA
The genomic DNA of *L. monocytogenes*was extracted from the cells by a commercial kit (Sigma, St. Louis, MO) according to the manufacturer’s instructions. Briefly, bacteria were cultured in BHI medium and collected by centrifugation. The cell pellets were lysed with a lysis solution, incubated for 20 min and then centrifuged. The supernatant was carefully collected and transferred into a new tube. Genomic DNA was extracted from the supernatant by binding the nucleic acids with magnetic beads. The extracted DNA was eluted in a solution, treated with RNase and washed with wash buffer.
